Naturally occurring protein has a role in chronic pain

HEIDELBERG, 12 June 2012 Researchers in France and Sweden have discovered how one of the body's own proteins is involved in generating chronic pain in rats. The results, which also suggest therapeutic interventions to alleviate long-lasting pain, are reported in The EMBO Journal.

Chronic pain is persistent and often difficult to treat. It is due, at least in part, to changes in molecular signalling events that take place in neurons, alterations that can ultimately disrupt the transmission of nerve signals from the spinal cord to the brain.

"We are fortunate to have a wide range of technologies that allow us to look more precisely at the molecular events that lead to the onset of chronic pain in animals," said Marc Landry, lead author of the study and Professor at the University of Bordeaux. "Our results show that the levels of the naturally occurring protein 14-3-3 zeta are higher in the spinal cord of rats that have chronic pain. Moreover, we have been able to demonstrate how 14-3-3 zeta triggers changes in the signalling pathway that leads to the symptoms of chronic pain."

The 14-3-3 zeta protein disrupts the interaction between the two subunits of the GABAB receptor, a protein complex found on the surface of nerve cells. GABAB receptors are G-protein coupled receptors, a family of receptors that regulate many physiological processes and which are frequently targeted for drug development.

The researchers used antibody labelling and microscopy techniques to investigate the molecular interactions of the signalling proteins. In cells and living animals, they were able to show that the 14-3-3 zeta protein interacts directly with the B1 subunit of the GABAB receptor. This interaction impairs the effective signalling of the receptor and limits the pain-relieving effects of the GABAB receptor under conditions of chronic pain.

The researchers also showed that the treatment of rats with a specific small interfering RNA (siRNA) or a competing peptide, molecules that interfere with the action of the 14-3-3 zeta protein, inhibited chronic pain.

"The impairment of the GABAB receptor by 14-3-3 zeta is a novel mechanism for the modulation of chronic pain," said Landry. "We see potential in combining the use of inhibitors that interfere with the action of 14-3-3 zeta together with existing drug treatments like Baclofen for chronic pain. Targeting the GABAB dissociation process may be of therapeutic interest since it may allow classical pain killers to be more effective."

I have to start out by saying that my family?doesn?t?really have any food allergies. But I love to cook and?bake, and I am always looking for healthier options to feed my family. I liked this cookbook, and I think?families that do have food allergies will love it.

Each kid-friendly recipe is broken down to let you know whether it is gluten-free, dairy-free, nut-free,?egg-free and soy-free. And there are many recipes that are free of them all. There are several sections?in the book called Cooking with Kids that give parents pointers on how to get their children involved in?the cooking and preparation process. There is even a whole section in the cookbook dedicated to party?food, which includes things such as chocolate cake, gluten-free cupcakes, corn dogs, and party mix.

We tried several of the recipes. My favorites were the Cheesy Stuffed Pizza Burgers, Sunset Pops, and?Indoor S?mores. I really liked the snack section of the cookbook, because I struggle with trying to find?my kids more nutritious snacks. The only downfall of some of the recipes were that they had ingredients?that I have never heard of. But I am sure if you are used to cooking for allergies, you would be familiar?with some of the ingredients. I also liked the fact that in many of the recipes you could substitute items?if you?weren?t?allergic to them. For instance, instead of using gluten-free chocolate chips in the Indoor?S?mores, I just used regular chocolate chips. The recipes are very versatile based on allergy needs.

Over the weekend, European countries agreed to lend up to $125 billion to?Spain?to help its crippled financial industry. Traders are concerned that it's only a temporary fix for the European debt crisis.

On Monday,?Italy?said that its economy contracted in the first quarter, raising fear that it may be the next country to ask for a bailout. The interest rates on?Italy's?10-year bond jumped to nearly 6 percent, a sign that investors are concerned about the country's finances.

The euro fell to $1.2498 in late trading Monday from $1.2507 late Friday. Before its drop, the euro rose as high as $1.2647 overnight. That was its highest point against the dollar since May 23.

Traders are also concerned about this weekend's Greek election. If anti-bailout politicians win, the country could be forced to leave the eurozone, a move that could hurt global financial markets.

The dollar was mixed against other currencies. The British pound rose to $1.5498 from $1.5462 and the dollar slipped to 79.44 Japanese yen from 79.48 yen.

But the dollar rose to 0.9609 Swiss franc from 0.9603 Swiss franc and to 1.0305 Canadian dollar from 1.0293 Canadian dollar.

Android A to Z: LG's L-series smartphones

This time on Android A to Z we're going to be talking about LG's L-series phones. They made a big splash earlier this year at Mobile World Congress, and a few carriers in Canada jumped at the chance to sell them, but then they seem to have fallen off the map. I'll admit, even I have to look and see what we're talking about when we have potential news about the L3, L5, or L7. Let's have a refresh.

Imagine LG's Prada phone, but remade for folks who don't buy Prada. The L-series phones are all stylish (and really resemble LG's iconic Prada phones -- check out our look at them in Barcelona) and have an emphasis on good looks. The entry-level L3 competes with phones like HTC's One V and Samsung's entry-level Galaxy phones. It's running Gingerbread on a 3.2-inch display, but it looks nice doing it. Compared to something like the Motorola Defy Mini, you get good looks along with your dirt-cheap pricing. The L5 takes things up a notch to Ice Cream Sandwich and a 4-inch screen, placing it in the middle of the road where most people look first when buying a new smartphone. They'll see the L5, and notice the build materials and design. Finally, the L7 looks to compete with the big dogs of the Android world, with all the bells and whistles you would expect from a high-end smartphone, in a damn fine looking external shell. All three are eye-grabbing, and getting people to notice is the first hurdle. We're really not sure why we don't hear more and see more about them, but we're guessing a saturated market has something to do with it. 

Maybe we'll see the L-series phones make their way into the spotlight, maybe we won't. But at least now we all know what we're talking about when we see them mentioned.

The Real Housewives of New Jersey Recap: True Love, True Lies

On this week's episode of The Real Housewives of New Jersey, "True Love, True Lies," we saw plenty of both as the gang partied both in Chicago and down at the Shore.

We break down all the love, laughs, and lies in THG's +/- recap!

Teresa is on the cover of yet another magazine?!? But really, is anyone shocked. If Teresa's in this for any reason it's certainly the fame…and the money. 

Plus 10 because the most amusing part is that no one can stop talking about the puppy. There on the cover of In Touch magazine is canine phobic Teresa with a cuddly pup. Of course it wasn't her idea.

The celebrity news magazine brought the adorable pooch along for the photo shoot. Maybe they were trying to soften Teresa's slightly tarnished image.

In the article Teresa claims her friends bully her because of her financial problems. Minus 8. Teresa never met a dollar she couldn't spend. It certainly doesn't look like she's buying the kid's clothes at Walmart. 

But who needs money when you've got love. Unfortunately we were treated to Teresa trying to give Joe a wake up call in Chicago which included a shot of the lube they used on their bedside table.

Did we really need to see that? Minus 15. Then Joe mentions giving himself his own wake up call. That's just way too much information.

As if that weren't enough we got a lovely glimpse into some romantic pillow talk between Joe and Teresa…

Joe: "Your breath smells ilke hot dogs from last night."
Teresa: "My breath smells like your hot dog."

If that doesn't make you feel all warm and fuzzy then I don't know what will. Minus 20 while I go find my toothbrush and try to burn that piece of dialogue from my brain.

But wait, it gets better. With rumors of Joe's cheating and their divorce swirling through the tabloids, Teresa has the perfect way fix things. The happy couple will be renewing their vows.

Plus 12. The Guidices can't even ring a doorbell without annoying people, I can't imagine how obnoxious their vow renewal ceremony will be. I'm laughing already and it hasn't even begun.

OK, let's move on to the actual real wedding which was absolutely beautiful. Plus 25. The ceremony was lovely and given the amount of sweat pouring through Jaime's purple shirt, I think he was having a good time at his reception. 

I'm hard pressed to choose a favorite moment but it might be a tie between the butterflies and the lighted lanterns. I just hope the floating balls of flame weren't a fire hazard. That would definitely ruin the romance.

Plus 10 because although I thought the mandatory hat rule was silly at first, it turned out to be a lot of fun. Who had your favorite hat? I'm voting for Jacqueline.

Back in Jersey the Gorgas were throwing a rockin' party at their shore home. Plus 8 because at least they had the decency to warn the neighbors. 

Before things got out of hand Rich witnessed Joe text his sister one more time asking for them to get help with their problems. Then in true Jersey style he got covered in whip cream and jumped in the pool.

Teresa Giudice, of course sees no reason for therapy and Plus 6 because now even Jacqueline is beginning to realize how warped Teresa truly is. 

Oh, and we wouldn't want to miss the moments on the bus to and from the wedding…

What was Chris thinking inviting everyone on the bus to Napa. That's one Hell of a road trip. Can anyone else see disaster looming ahead?

And to close out the entire affair we have the stirring words of Joe Guidice as he yells out to his wife…"Tre, your tits are hanging out!"

Minus 13. Here's to keeping it classy with the folks from Jersey.

Lowest Interest Rates personal loan schemes for Property Loans - SBI

SBI has full competition head on by launching a personal loan scheme against property loans at interest rates lower than some private banks.

The scheme, aimed at existing home loan borrowers and linked to the price of property, is offering personal loan at 11.25 % to existing home loan customers.

The offer is related to the top-up scheme of ICICI Bank at 12.50 % and HDFC, which have hooked their loans at 12.25 %. The new product, called 'SBI Home Equity scheme', gains relevance in the context of the rise in property prices over the last few years.

The personal loan entitlement under the scheme is linked to the value the property for which the borrower is already paying EMI. The lender will give a maximum 75 % of the value of the property after deducting the outstanding amount of the loan.

The interest rate of 11.25 % charged by SBI is benchmarked to the bank's base rate of 10 %.

Similarly, rates offered by other banks like ICICI Bank and HDFC for top-up loans are benchmarked to their base rates or prime lending rates. The scheme, which links home loans to personal loans, will attract home loan customers of other banks.

Customers may shift their loan account to SBI, particularly in the context of the Reserve Bank of India banning banks from charging any prepayment penalty. SBI is also offering overdraft facility on the new scheme at 11.50 %.

SBI has emerged as one of the largest players in the home loan market with a portfolio of more than Rs 1 lakh crore.

Renewable energy's growing pains

Renewable energy received record investment in 2011 and expanded massively, but it also struggled with dwindling political support and plummeting prices. The industry faces several more years of growing pains before it can properly compete with fossil fuels.

By the end of 2011, the global power capacity from renewables was more than 1360 gigawatts, and renewables supplied 20.3 per cent of global electricity, according to the REN21 Renewables 2012 Global Status Report.

Meanwhile, investment in renewables increased by 17 per cent last year to a record $257 billion ? six times what it was in 2004, according to a report by the UN Environment Programme. The UNEP report, Global Trends in Renewable Energy Investment 2012, concludes there were particularly big gains for solar power, which received $147 billion ? 52 per cent more than in 2010.

Yet at the same time, solar power companies suffered massive drops in share prices. Six major companies, including Solyndra and Solar Millennium, have sought bankruptcy protection.

The main reason is the steep drop in the cost of solar panels over the last three years, largely due to a switch to large-scale manufacturing. Solar power is now cheaper than diesel in countries such as India. Many governments, including the UK, also cut their financial support for solar, as tumbling prices meant consumers did not need such large subsidies to buy a panel.

It is normal for weaker firms to fall by the wayside as industries ramp up, says Michael Liebreich, chief executive of Bloomberg New Energy Finance.

"The challenge for policy-makers is to reduce support mechanisms at just the right pace," he says. Cutting subsidies too fast will stop renewables in their tracks, but maintaining them for too long will be a waste of money.

One?s sexual life problems are generally influenced by several factors such as psychological and physical. Those who suffer from certain diseases like diabetes has a greater risk of facing problems of sexual life.

So, how diabetes can affect your sexual life or vice versa? Research shows, twenty-five percent of all women with diabetes and about 50 percent of men will experience some kind of sexual problems or loss of sexual desire as a result of their condition.

In Men
Long-term diabetes in men can cause damage to the nervous system, which is involved in the complex process of erection. This means that men with diabetes may suffer from erectile dysfunction (ED) and are unable to obtain or maintain an erection. About one third of men with diabetes will experience impotence at the end.

In fact, several new men know they have diabetes when they seek treatment of erectile dysfunction related to them. But you need not worry, this can be overcome by setting the proper diet and supported by the consumption of drugs or insulin.

In Women
A number of women with diabetes may suffer from recurrent vaginitis (vaginal inflammation), which is usually caused by fungal infections. This condition will make sex uncomfortable and painful. Even in some women, may experience itching or burning sensation, and a white discharge.

Women with diabetes also get recurrent cystitis ? a disease of Urinary Tract Infection (UTI). In addition, some evidence suggests that women with diabetes tend to have problems with arousal, and the clitoris (the nerve center of the female sex) may not respond to stimuli in the usual way.

Is it possible to have sex after suffering from diabetes?
If you feel diabetes cause problems with your sex life, immediately consult with dokter.Biasanya, the doctor will try to find out whether sexual problems are caused by defects in the nervous or circulatory system due to diabetes , or whether it is more due to psychological factors. Often these differences are difficult to group them.

In any case, when the problem of sexual dysfunction began to occur regularly and are likely to deteriorate, so will the psychological factors involved in it.

Men who have diabetes and impotence can be helped with medication prescribed by doctors such as sildenafil (Viagra ), tadalafil (Cialis) and vardenafil (Levitra). However, if the test results indicate that your nervous system has not been damaged by diabetes, and no circulatory problems are related, then you really do not have problems with their sex lives. It takes the support of partners and doctors to cope with this condition.

Happily, the number of diapers that we go through per month has been drastically reduced since the babies first came home. These days, we go through roughly?600 daytime diapers and 140 nighttime diapers per month. That's still a lot of diapers!

This is most certainly not the blog to read for regular deal updates, but this is one deal that I absolutely must pass on!

Walgreens is having their buy-one-get-one free deal on diapers right now! It comes at a very VERY good time for our family since we just ran out of baby shower diapers in the last couple of months. Many thanks to my fellow quad mom, Jamie, for tipping me off that now is the time to stock up!

The deal is pretty simple (thank goodness!):

All Walgreen Brand Jumbo Diaper Packs are 2/$8.99. They are regularly 1/$8.99.

The problem with these types of deals is that most Walgreens only stock a few packages of diapers in each size. Before kids, I used to play the drug store game. When I found something I wanted to stock up on, I would drive around to a bunch of different stores until I found enough of what I was looking for.

These days, I don't have that kind of time. So this afternoon, I drove to my favorite Walgreens (yes, I do have a favorite Walgreens), and asked them to order 44 packages of Size 3 diapers. The manager was extremely?accommodating and should have them in the store by late Friday afternoon so that I can pick them up that evening. It's worth asking at your local Walgreens if you are in need of diapers!

I ordered 44 packages of diapers which is enough to get us close to three months worth of diaper wearing. From what I understand, these deals tend to be on three month cycles with Walgreens. So stock up if you have babies, are pregnant, or are planning to go to a lot of baby showers soon! :)

We are currently subscribed to Amazon Mom which is the?cheapest way that I can find to buy diapers without doing reward clubs and other things like that that require more brain cells than I have to spare at the moment. Since I'm stocking up on diapers this week, I'll be skipping my shipment from Amazon for the next few months.

These easy deals are definitely the type of shopping that I like to do these days!

Note: The Walgreens website is only showing Size 4 and up diapers at the 2/$8.99 price. However, all Walgreens Brand Jumbo packs are 2/$8.99 in stores! And this is a national deal!

Arctic getting greener

Recent years' warming in the Arctic has caused local changes in vegetation, reveals new research by biologists from the University of Gothenburg, Sweden, and elsewhere published in the prestigious journals Nature Climate Change and Ecology Letters.

The results show that most plants in the Arctic have grown taller, and the proportion of bare ground has decreased. Above all, there has been an increase in evergreen shrubs.

"We've managed to link the vegetation changes observed at the different sites to the degree of local warming," explains researcher and biologist Robert Bj?rk from the University of Gothenburg.

Shrubs and plants more widespread

Comparisons show that the prevalence of vascular species, such as shrubs and plants, is increasing as temperatures rise. The degree of change depends on climate zone, soil moisture and the presence of permafrost.

Researchers working on the International Tundra Experiment (ITEX) have been gathering data for almost 30 years. By analysing changes in vegetation in 158 plant communities at 46 locations across the Arctic between 1980 and 2010, they have been able to identify a number of general trends.

"We've managed to show that the vegetation changes in our fixed plots are a result of local warming at numerous sites across the world's tundra," Robert Bj?rk says.

Summer temperatures and soil moisture implicated

ITEX was started up in the USA in 1990 when agreement was reached on a joint manual with standardised protocols which have since been used throughout the Arctic.

"The response of different plant groups to rising temperatures often varied with summer ambient temperature, soil moisture content and experimental duration, with shrubs expanding with warming only where the ambient temperature was already high, and grasses expanding mostly in the coldest areas studied," explains Ulf Molau, professor of plant ecology at the University of Gothenburg and for many years a member of the Intergovernmental Panel on Climate Change (IPCC).

Major changes

The results indicate strong regional variation in the response of tundra vegetation to rising temperatures.

"This means that particularly sensitive regions following the combined effects of long-term warming in the Arctic may see much greater changes than we have observed to date," Ulf Molau says.

This is a timely insight now that Sweden, as chair of the Arctic Council in 2011-13, has prime responsibility for producing the Arctic Resilience Report. Experience from ITEX will also be used in the next IPCC assessment report in 2014.

Ex-Apple engineer reportedly buying NBA's Memphis Grizzlies

Apple's hardware seems to have been updated today, so I hear.

However, one man who might once have thought he'd be involved in those updates has reportedly achieved a slightly more exalted goal: he has bought himself an NBA team.

Robert Pera, 34, once an Apple hardware engineer and now the founder and CEO of Ubiquiti Networks, a company that seeks to provide Web connectivity cheaply for emerging markets, is reportedly buying the Memphis Grizzlies.

The way ESPN shoots it, Pera is an avid, nay, obsessive basketball player who owns 64 percent of Ubiquiti, a company that has its homes in both Taiwan and San Jose, Calif.

Naturally, those whose hearts contain cynical hardwood will speculate, on seeing the words "San" and "Jose," that Pera intends to ship the team away from Memphis.

More Technically Incorrect

ESPN insists, though, that there are nasty financial clauses preventing such a move before 2021. Moreover, the Grizzlies current owner, Michael Heisley, says he ended talks with Oracle's Larry Ellison precisely because Ellison wanted to spirit the team to other climes.

Still, some may remember what seemed to be firm promises made by the new owners of the Seattle Sonics, before they thundered the team out of town to the basketball, um, hotbed of Oklahoma City.

Pera, who left Apple when he was 25, has reportedly found a mere $350 million to buy the Grizzlies. It helps being on Forbes' fine, but somewhat exclusive, list of billionaires.

The Grizzlies are a likeable young team that last year made the playoffs, even defeating the San Antonio Spurs -- and this year again made the playoffs before somehow allowing the Los Angeles Clippers to bamboozle them.

One wonders whether Pera will follow the technological path blazed by the Dallas Mavericks' owner Mark Cuban, who has proved to be one of the best -- as well as one of the most pleasantly vociferous -- owners in the NBA.

HBT: Oswalt moving from Triple-A to Double-A

Roy Oswalt?s second minor-league start involved giving up a grand slam to Royals stud prospect Wil Myers on the way to allowing five runs in four innings at Triple-A.

He?ll move down a level to Double-A for his third tune-up outing tomorrow night and if things go well he could be on the verge of joining the Rangers? rotation, especially with Derek Holland on the disabled list and fill-in Alexi Ogando now sidelined by a groin injury.
